TransFix®: A Cellular Antigen Stabilisation Reagent


TransFix® is a patented, CE marked cellular stabilisation solution that preserves cell surface antigens for a minimum of 10 days at 2-22°C and for 4 days at 37°C (1). TransFix is added to whole blood samples at the time of venepuncture, and provides stabilisation so that extended storage and/or transportation can be achieved thus allowing greater flexibility for testing and potential reduction in costs.
TransFix® is used to stabilise whole blood for flow cytometry and blood donor screening, plus Cerebrospinal Fluid (CSF) (2), Bone Marrow and Fine Needle Aspirates. It is available in several formats including blood or CSF sample storage tubes, direct draw vacuum blood collection tubes, and bulk volumes.

Sample StabilityWhole blood degrades over time and blood samples older than 36 hours are no longer suitable for flow cytometric examination. Addition of TransFix® to blood samples at the time of collection has been shown to significantly extend the integrity of the samples. The immunophenotypic profile of markers including CD3, CD4, CD8, and CD45 is maintained for 10 days.
Sample Transport & BatchingThe use of TransFix® is beneficial in situations where sample storage, batching or transportation is required prior to analysis. This may be particularly relevant in resource poor or remote areas of the world where there may be extended transit times between the phlebotomy site and laboratory. TransFix® treated samples may also be batched together before transporting, thereby reducing the number of shipments to the laboratory and thus reducing costs. In addition, the batching of samples for testing may result in reduced laboratory staffing costs.
TransFix® also reduces the need to recall patients for additional phlebotomy by extending the window of opportunity for sample testing, resulting in significant cost savings. This may be useful in circumstances where unexpected delays in testing have occurred, e.g. machine break down or transportation delay. TransFix® may also reduce the need for out of hours testing as treated samples can be stored until normal working hours. The use of TransFix® may therefore reduce the costs associated with out-of-hours testing.
Easy To UseSamples should be treated with TransFix® as soon as possible after phlebotomy. Samples are added to TransFix® containing tubes and simply inverted and then stored at 2-8°C. The sample can then be treated as a “routine” preparation for subsequent analysis. The dilution factor must be taken into account when analysing absolute count data.

NAMPT [VISFATIN/PBEF] – Linking NAD Biology, Metabolism and Cancer

Nicotinamide phosphoribosyltransferase (Nampt; pre-B cell colony-enhancing factor; PBEF; Visfatin) is an 52kDA adipokine secreted by adipose tissue and involved in the biosynthesis of nicotinamide adenine dinucleotide (NAD+). Two forms of Nampt exist, an intracellular form (iNampt) and an extracellular form (eNampt). While the function of iNampt as an essential and rate-limiting NAD+ biosynthetic enzyme is well established, the physiological role of eNampt is still a matter of debate. It has been shown that Nampt expression is increased in different diseases, like diabetes, obesity, fetal growth retardation, sepsis, inflammatory bowel disease, Crohn’s disease and rheumatoid arthritis. Furthermore, Nampt might be a marker of pre-eclampsia and of endothelial health.

Runx2 - a key transcription factor associated with osteoblast differentiation

Runx2, is an essential transcription factor of skeletal tissues that is involved in the regulation of osteoblast differentiation and bone formation. Runx2-null mice have neither bone tissue nor osteoblasts. FGF receptor signaling, TGF-β, and BMP all activate transcription of Runx2, resulting in inhibition of myogenesis and myogenic differentiation.

BMP-induced Runx2 cooperates with BMP-activated Smads to induce osteogenesis. Runx2 has also been shown to play a role in the regulation of chondrocyte hypertrophy and tooth eruption.

Samples Available For Blood Controls For Flow Cytometry


The quantitation of CD4+ cells by flow cytometry is a vital tool in evaluating human immunodeficiencies. Cytomark have developed two stabilised preparations of human blood with known lymphocyte absolute counts including CD4+ cells. They are intended for use as daily run controls in blood sample analysis to ensure that sample processing, equipment and operators are all performing optimally thereby providing quality assurance in the immune monitoring of patients.

CD4 Blood Controls are available in two formats; a normal CD4 blood cell profile - >700 cells/µl and a low CD4 blood cell profile - between 200 and 300 cells/µl. The low control simulates CD4 levels observed in advanced HIV/AIDS patients. The CD4 blood controls can be purchased separately or as a pair.

Non-lytic Ig-based Chimeric Fusion Cytokines with Long Circulating Half-life
Unfortunately, most cytokines have short circulating half-lives. Therefore, to facilitate the study of cytokine effects in vivo, a variety of non-lytic immunoglobulin-based chimeric cytokine fusion proteins have been created, in which a cytokine sequence had been genetically fused to the hinge, CH2 and CH3 regions of an immunoglobulin.

These non-lytic fusion proteins possess both the biological functions of the cytokine moiety and a prolonged circulating half-life determined by the Fc domain. They retain the potential to direct immune cytolytic mechanisms, antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC) against cellular targets bound by the amino terminal binding moiety. These fusion molecules also have the promise of being minimally to negligibly immunogenic since they are made entirely from elements derived from the species to be treated.

ELISA Kit for Galectin-9 (GAL9)


Galectin-9 (Gal-9) is a member of animal lectins that have an affinity to beta-galactosides. Gal-9 was first cloned from tumor tissues of Hodgkin’s disease that is frequently characterized by eosinophilia. Galectins exhibit various biological functions, such as cell aggregation and adhesion, proliferation,cell death, modulation of inflammation and metastasis.First, Gal-9/ecalectin was identified as a potent T cellderived eosinophil chemoattractant (ECA).

Gal-9 exhibits ECA activity in vitro and in vivo, activates eosinophils, and influences eosinophil survival. It requires divalent galactoside- binding activity but not its linker peptide to exhibit its functions , and some matrix metalloproteinases are involved in the release.

Immunology

Interleukin 18 (IL-18) is an 18 kDa novel cytokine identified as a co-stimulatory factor for production of interferon-γ (IFN-γ) in response to toxic shock. IL-18 acts on T helper type 1 (Th1) cells in combination with IL-12. It strongly induces production of IFN-γ by these cells. IFN-γ is produced by activated T and NK cells and play critical roles in the defence against microbial pathogens. IFN-γ activates macrophages, enhances NK activity and B cell maturation, proliferation and Ig secretion, induces MHC class I and II antigen expression, and inhibits osteoclast activation.
Regulatory T cells (Treg) suppress overactive immune responses by inhibiting the function of activated T cells. These cells are very important for immunological tolerance of self antigens.
Natural Killer T (NKT) cells, a type of T cell that plays a significant role in immune response, produces a large quantity of INF- and IL-4 in response to glycolipids that presented by CD1d molecules. In recent years, NKT cells are reported to play a part in diabetes and tumor immunity. Therefore, a technology allowing quantitative measurement of CD1d-positive NKT cells would be a useful tool for immunology and clinical laboratory examinations.
The development of MHC Tetramer technology has provided a breakthrough in the ability to follow T cell populations defined by their antigen specificity. Tetramers have been used widely to obtain a detailed analysis of the distribution and frequency of conventional CD4+ and CD8+ antigen-specific T cells during a variety of immune responses.

ELISA Kit for Interleukin 35 (IL-35)

Interleukin 35 (IL-35) is an IL-12 family cytokine that is produced by regulatory, but not effector, T cells and plays a role in immune suppression. It is a dimeric protein composed of IL-12α and IL-27β chains, which are encoded by two separate genes called IL12A and EBI3, respectively.

Secreted by regulatory T cells (Tregs), IL-35 suppresses inflammatory responses of immune cells. Studies in mice show the absence of either IL-35 chain from regulatory Tregs reduces the cells ability to suppress inflammation; this has been observed during cell culture experiments and using an experimental model for inflammatory bowel disease. To produce its suppressive effects, IL-35 has selective activities on different T cell subsets; it induces proliferation of Treg cell populations but reduces activity of Th17 cell populations.
