46 Multiplex Tyrosine Capture ELISA Assay

Receptor tyrosine kinases (RTKs) play important roles in converting signals from extracellular to intracellular.  Tyrosine autophosphorylation generates recruitment sites for downstream signalling proteins containing Src homology-2 (SH2). Binding of different SH2 domain-containing signalling proteins represents the activation of different signalling pathways. The SH2 domain-based RTK profiling kit from Signosis is an ELISA-like assay, allowing the simultaneous measurement of 46 SH2 domain bindings using either cell lysate or peptides.  This allows the activation of signalling pathways to be assessed through quantitative examination of SH2 domain binding. 


  1. Multiplex - A single assay measures the interaction of RTK with 46 SH2 domains.
  2. Quantitative - The difference between individual proteins in the interaction of the SH2 domains can be quantitatively compared. 
  3. Simple - The assay is performed using the same procedure as an ELISA.

ELISA Kit for Hepcidin (Hepc)

Hepcidin is a peptide hormone produced by the liver discovered in 2000, that appears to be the master regulator of iron homeostasis in humans and other mammals. In humans, HAMP is the gene that encodes for hepcidin.

Hepcidin preprohormone, prohormone and hormone size are 84, 60 and 25 amino acids respectively. 20 and 22 amino acid forms of Hepcidin also exist in the urine. The N terminal region is required for function, and deletion of 5 N-terminal peptides results in a loss of function. Hepcidin has a beta-sheet rich structure with 4 disulfide bonds. Hepcidin directly inhibits ferroportin, a protein that transports iron out of cells that store it. This prevents enterocytes of the intestines from secreting iron into the hepatic portal system, reducing iron absorption.

PCSK9 : an attractive drug target for lowering LDL-C?

PCSK9 plays a major role in cholesterol metabolism through posttranslational down regulation of LDLR, the receptor responsible for clearing LDL-C from plasma.
PCSK9 (neural apoptosis-regulated convertase, NARC-1) is a 692-residue extracellular protein representing the 9th member of the secretory subtilase family expressed primarily in the kidneys, liver and intestines.

Genetic studies mapped PCSK9 along with LDLR and APOB to cause autosomal dominant hypercholesterolemia (ADH). Gain-of-function mutations increased plasma levels of low-density lipoprotein cholesterol (LDL-C), whereas nonsense or missense (loss-of-function) mutations, which interfere with folding or secretion of PCSK9, led to a reduction of plasma levels of LDL-C and an 88% decrease in the risk of coronary heart disease (CHD).
These findings led to a hypothesis that PCSK9 exerts its role in cholesterol metabolism through posttranslational down-regulation of LDLR, the receptor responsible for clearing plasma LDL-C. The hypothesis is consistent with a secreted form of PCSK9 bound directly to the first epidermal growth factor-like repeat (EGF-A) of LDLR and resulting in degradation of the receptor. PCSK9 appears to be an attractive drug target for lowering LDL-C.

Isotype Control Antibodies

Isotype controls are most commonly used in flow cytometry experiments for delineating the non-specific background signals caused by the primary antibodies. Isotype control antibodies are designed to retain all the non-specific characteristics of the primary antibodies, except the specificity for target cells in the experiment. The purpose is to confirm the binding of the primary antibody with the target cell is specific and not a result of other non-specific bindings and protein interactions. Thus, isotype controls need to be matched exactly with the primary antibodies to the host species and the isotypes, including heavy chain and light chain. There are many immunoglobulin subclasses available for isotype controls. The isotype control immunoglobulin(Ig) subclasses are IgG1,IgG2a, IgG2b, IgG3 and IgM. Most isotype controls are monoclonal. However, when it comes to more than one IgG subclass, they are not suitable. Therefore, Abnova also offers several isotype control polyclonal antibodies for use inthese circumstances.

Abnova carries conjugated isotype control antibodies, and unlabeled monoclonal antibodies. The conjugation formats and staining conditions are also required to be identical in the experiment in order to accurately assess the level of specific binding. Choosing a conjugated antibody can save you a lot of time and efforts. However, as an alternative, choosing an unlabeled antibody will give you more flexibility for your experiment if you are using more than one dye.

RIP Assay Kits for the Immunoprecipitation of Ribonucleoproteins from Cell Extracts

MBL have developed two RIP assay kits which enable the immunoprecipitation of mRNA-RBP (RNA binding protein) complexes with RBP specific antibodies.
RIP-Chip works on the same principle as the widely used ChIP-chip. It is the immunoprecipitatation of ribonucleoproteins (RNP) from cell extracts using an antibody raised against the RBP of interest. This procedure is then followed by microarray analysis.
