46 Multiplex Tyrosine Capture ELISA Assay

Receptor tyrosine kinases (RTKs) play important roles in converting signals from extracellular to intracellular.  Tyrosine autophosphorylation generates recruitment sites for downstream signalling proteins containing Src homology-2 (SH2). Binding of different SH2 domain-containing signalling proteins represents the activation of different signalling pathways. The SH2 domain-based RTK profiling kit from Signosis is an ELISA-like assay, allowing the simultaneous measurement of 46 SH2 domain bindings using either cell lysate or peptides.  This allows the activation of signalling pathways to be assessed through quantitative examination of SH2 domain binding.

    More recently new technologies have been introduced that allow the testing of multiple biomarkers in a single assay. Doing this with immunoassays is difficult and the level of cross-talk between individual assays within the multiplex are difficult to measure and often lead to significant variation in results between labs and over time.
