FIX & PERM® by An Der Grub

An Der Grub’s FIX&PERM® for suspension stainings and flow cytometric analysis of intracellular antigens.

Flow cytometric analysis with monoclonal antibodies has been primarily restricted to cell surface molecules. Intracellular structures including cytoplasmic or nuclear enzymes, oncoproteins, cytokines and immunoglobulins were largely excluded from such studies.

Also excluded from flow cytometric studies were cytoplasmic localizations of well established membrane molecules like CD3 and CD22, which, in their cytoplasmic form, are the most reliable lineage markers in undifferentiated leukemia. With the FIX&PERM® Kit flow cytometric analysis of intracellular antigens has become as easy as surface antigen studies.

The FIX&PERM® Cell Permeabilization Kit contains 2 reagents: Fixation Medium (Solution A) and Permeabilization Medium (Solution B). It is intended for first fixing cells in suspension with Solution A and then permeabilizing the cell membranes with Solution B. This procedure gives antibodies access to intracellular structures and leaves the morphological scatter characteristics of cells intact.

Specific formulations reduce background staining and allow simultaneous addition of permeabilization medium and fluorochrome labeled antibodies. FIX&PERM® is suitable for the analysis of normal and malignant leukocyte populations derived from various human biological samples (blood, bone marrow and others) using flow cytometry.

Sirtuin 1 (Intracellular) ELISA kit

Sirtuins (Silent information regulators; SIRTs) deacetylate and/or ADP-ribosylate lysine residues of target proteins in an NAD+-dependent manner. Sirtuins are critical regulators of many cellular processes, including insulin secretion, the cell cycle and apoptosis. Sirtuins, in particular SIRT1, emerged as critical regulators of longevity and aging in several model organisms. Furthermore, a variety of age-associated diseases such as type 2 diabetes, obesity, osteoporosis, and Alzheimer’s disease are associated with sirtuins. SIRT1 has been suggested as potential biomarker for tumorigenesis.

This Adipogen ELISA comes in 2 pack sizes: 1 x 96 well plate and 2 x 96 well plates.

AdipoGen – now distributed by Caltag Medsystems.

Adipogen supply advanced ELISA kits for the life science reagents market including the areas of cancer, immunology, inflammation, metabolic syndrome (diabetes, obesity), stem cells and neurodegeneration.
In addition to the antibody and protein facilities, AdipoGen also own chemical laboratories, enabling them to develop new and innovative biochemicals.

The AdipoGen Institute for Life Science connects immunology to metabolism. In the next decades both diseases, obesity and diabetes, are considered to increase significantly worldwide. It becomes more and more evident that both diseases are not only a result of unbalanced nutrition but are closely related to inflammatory processes. To establish better cures of the diseases, it is necessary to understand the cross-talk between the hormones and cytokines, which are secreted from fat cells and immune cells, and to understand the principles of the specific binding of ligands to the receptors. The Adipogen Institute for Life Science offers a drug design platform, through i) discovering and characterizing adipokines and cytokines, which are secreted from fat cells and immune cells regulating glucose and fat metabolism, and ii) identifying and characterizing the receptor genes of these secreted factors.

Abnova offer wide range of kits!

Abnova have over 1000 kits on their catalogue and here at Caltag Medsystems we supply them all!

This week we’re talking about ELISA kits.
Enzyme-Linked Immunosorbent Assay (ELISA) kits are used mainly to detect and quantify proteins, antibodies, peptides, or hormones in a sample. In ELISA’s, antigens are immobilized on a solid support, either coated directly or through specific capture antibodies. Primary detection antibodies are then applied, forming a complex with the antigen. If conjugated with an enzyme, the detection antibody can directly be used to quantify the antigen, or can itself be quantified by another enzyme-conjugated secondary antibody. The method chosen depends on which target you are investigating.
Featured Abnova kits this month are the following:
  • Corticosterone ELISA Kit (product code: KA0468)

Monosan announce the release of HGAL for the detection of follicular lymphoma.

This RUO antibody is available as concentrate or predilute (ready to use). Monosan’s HGAL is compatible with most automated staining platforms.

HGAL is a mouse monoclonal antibody that has the highest sensitivity of all established diagnostic markers (CD10, bcl6, and bcl2) for follicular lymphoma. It marks both follicular components and the diffuse interfollicular components that other markers are not sensitive enough to detect. CD10 and bcl6 have been more difficult antibodies to work with on lymphoid tissues, but HGAL doesn’t share this laborious characteristic. The high degree of specificity for germinal center B-cells makes anti-HGAL an ideal marker for the detection of germinal center-derived B-cell lymphomas.
